[Indications]

HR-positive, HER2-negative advanced/metastatic breast cancer with letrozole (first-line) or fulvestrant. 75mg is dose-reduction strength from 125mg.

[Mechanism of Action]

Highly selective CDK4/6 inhibitor. Prevents Rb phosphorylation, arresting G1-S cell cycle transition and inhibiting tumor proliferation.

[Dosage]

125mg starting dose daily 21 days on/7 off per 28-day cycle. Reduce to 100mg then 75mg for toxicity. Take with food.

[Adverse Reactions]

Neutropenia (75%), fatigue, nausea, stomatitis, alopecia, infections. Febrile neutropenia, pulmonary embolism, ILD.
