[Indications]

1. First-line EGFR mutation-positive advanced NSCLC. 2. Squamous NSCLC after platinum chemotherapy. 3. NSCLC with uncommon EGFR mutations (G719X, L861Q, S768I).

[Mechanism of Action]

Second-gen irreversible ErbB family blocker covalently binding EGFR/HER2/ErbB4. Superior to first-gen reversible TKIs with broader inhibition and OS benefit.

[Dosage and Administration]

Starting dose: 40mg. 30mg for dose reduction. Take on empty stomach 1h before or 3h after meal. Dose escalate to 50mg if tolerated.

[Adverse Reactions]

Diarrhea (95%), rash (89%), stomatitis (71%), paronychia (56%). ILD, hepatotoxicity, keratitis, LV dysfunction.
