[Mechanism of Action]

Second-gen irreversible covalent ErbB family (EGFR/HER2/HER4) inhibitor. ARCHER 1050: superior OS vs gefitinib in first-line EGFR-mutant NSCLC.

[Dosage]

Starting dose: 45mg daily. Reduce to 30mg then 15mg for toxicity. Prophylactic loperamide and skin care recommended.

[Adverse Reactions]

Diarrhea (87%), rash (78%), paronychia (62%), stomatitis (44%). ILD (fatal), severe diarrhea, hepatotoxicity.
